Engineered Bacteria Can Overthrow Tumors During Cancer Immunotherapy

The genetic programming help engineers create new systems that can respond and sense the changing environment followed by developing an effective solution compared to the present molecular-based therapeutics. In the cancer immunotherapy, the immune defenses itself are used to combat cancer and this method has helped transform cancer treatment; however, only a handful of it respond to the treatment and the systemic therapy generally cause a lot of side effects. The development of therapies that can stimulate an effective, anti-tumor immune response in a solid tumor exclusive of activating systemic toxicity is the significant challenge. Columbia University Irving Medical Center (CUIMC) and Columbia Engineering scientists have engineered a strain of non-pathogenic bacteria that can address this challenge by colonizing in solid tumors and safely transfer powerful immunotherapies that imitate a Trojan horse that can cure tumors from within.

The therapy helped seize tumor regression and also control uninjected tumor lesions. The bacterial cancer therapy is referred to as the abscopal effect by the researchers. The engineered bacteria can target the tumors by stimulating the immune system to control tumors metastases, which are small to be detected using imaging or other approaches. The bacteria such as non-pathogenic E. coli could be used as it released better therapeutic effect and caused less of havoc anywhere else in the body.

For programming the bacteria, the researchers chose to release a nanobody that targets CD47protein. The tumors after the bacterial invasion experience inflammation and ingestion that ultimately activates the T cells for tumor necrosis. The priming of tumor-specific T cells by the bacteria in the tumor can help treat distant tumors. Researchers at the Royal College of Surgeons develop a probe that can light up in the presence of tumors and make it easier for the surgeons to spot and destroy them. This new tool can help improve surgical outcomes by providing them real-time and informative images.
